He enrolled at the University of Wisconsin-Madison, but his studies were interrupted by World War II. He enlisted in the U.S. Army Air Corps in 1943 and served as a bomber pilot in Europe. He flew a total of 30 combat missions and was awarded the Distinguished Flying Cross.

In 1959, he was selected as one of the original seven astronauts for the NASA space program. He made his first space flight in 1971 on the Apollo-Soyuz Test Project, which was the first joint U.S.-Soviet space mission. He retired from NASA in 1977 and from the Air Force in 1979.
In 1982, he founded the Slayton Aerospace Company, which provided consulting services to the aerospace industry. He also wrote an autobiography, entitled Deke!, which was published in 1988.
