Deedee Magno was born on April 2, 1975 in the Philippines. Her parents are of Filipino, Chinese, and Spanish descent. She has two older sisters. Magno was raised in the Philippines until the age of six, when her family moved to San Diego, California.

Magno began her career as a child actress, appearing in various commercials and television shows. In 1992, she made her film debut in the movie Encino Man. She also had a recurring role on the television show Blossom.
In 1996, Magno auditioned for the role of Pearl on the television show Steven Universe. She was cast in the role and has voiced the character since the show’s debut in 2013. Magno has also voiced the character of Connie Maheswaran on the show.
Magno has appeared in a number of stage productions, including Rent and Avenue Q. In 2015, she made her Broadway debut in the musical Fun Home.
Magno is married to actor Jason Wong. The couple has two children together.
