Born in Paisley, Scotland, on January 2, 1969, McGillion was raised in a working-class family. His father was a welder and his mother was a homemaker. He has two older sisters.
McGillion attended the University of Glasgow, where he studied English literature. He began his acting career in the early 1990s, appearing in various stage productions. His first film role was in the 1995 independent film Trainspotting.
In 2004, McGillion landed the role of Dr. Carson Beckett on Stargate Atlantis. He appeared in all 100 episodes of the series, which ran for five seasons.
Since Stargate Atlantis ended in 2009, McGillion has had roles in several television series, including Supernatural, NCIS, and The Flash. He has also appeared in the films Saw 3D (2010) and Oculus (2013).
