Born in Detroit, Michigan, on November 21, 1937, Thomas is the eldest child of comedian Danny Thomas and his wife, the former Rose Marie Cassaniti. Her sister, Terre Thomas, is a television producer. She has two brothers: Tony Thomas, a television and film producer, and Daniel Thomas, a businessman.
Thomas was educated at the Immaculate Heart High School in Los Angeles and the University of Southern California, from which she graduated in 1959 with a bachelor’s degree in English.
Thomas’s first acting role was on her father’s sitcom, Make Room for Daddy (1953–57), on which she appeared sporadically as a child. After graduating from college, she appeared in episodes of The Donna Reed Show (1958–66) and The Virginian (1962–71). In 1966, she was cast as the title character in the ABC sitcom That Girl, which ran for five seasons.
Thomas’s other television credits include guest roles on The Love Boat (1977–86), Murphy Brown (1988–98), Friends (1994–2004), and Law & Order: Special Victims Unit (1999–present).
